Overview
MOTS-c is a 16-amino-acid peptide (sequence MRWQEMGYIFYPRKLR) encoded within the mitochondrial 12S rRNA gene, a genomic region previously believed to code only for oxidative-phosphorylation components. First described by Lee et al. in 2015, it is considered a founding member of the mitochondrial-derived peptide (MDP) class: molecules that appear to act as retrograde signals carrying information from mitochondria to the nucleus and other cellular compartments.
Published research has characterized MOTS-c's translocation to the nucleus under conditions of metabolic stress, where it is reported to influence gene-expression programs tied to glucose uptake, fatty-acid oxidation, and thermogenesis, largely through interaction with the AMPK energy-sensing pathway. This mechanism has made it a subject of laboratory research into metabolic regulation and exercise physiology.
